No, you should be ok.

I'm on book 3 in the first omnibus, so I'll be starting this soon. Each book (so far) has filled in enough background info to not be completely lost. If you hadn't read the earlier books (and short stories) there'd be some references that might not make as much sense, but you wouldn't be completely lost (especially if you're already familiar with the W40K universe, which I wasn't.)
