    Will this work to turn on the heating to one room but leave the rest of the radiators off (I am assuming that's not how they work? As they're in line but I'm hoping so as I have one really cold room!)

    Yes - exactly as you describe, assuming the thermostat is on for the boiler and the boiler is on.

    If you have one particularly cold room which fails to get warm, this will keep the radiator open whilst the others have closed.
    It is likely a balancing issue though - if you turn down the lockshield valves down all the way, open them up a quarter of a turn and then monitor the temps and then keep adjusting them a little more open until all rooms hit the temps at the same time. (edited)
